Agonist paradox

A GnRH agonist first stimulates luteinizing hormone and follicle-stimulating hormone release, then, with sustained exposure, downregulates and desensitizes pituitary receptors. The net effect after the initial flare is reduced sex-hormone output, which underlies its clinical uses.
