Chimeric pro-apoptotic peptidomimetic (investigational)

Adipotide

Adipotide (Prohibitin-TP01) is an experimental chimeric peptidomimetic that pairs a homing sequence targeting adipose-tissue blood vessels with a pro-apoptotic motif. Studied in animal obesity models, its clinical development was discontinued. It is not an approved therapeutic in Canada.

Overview

Adipotide (Prohibitin-TP01) is an experimental chimeric peptidomimetic that pairs a homing sequence targeting adipose-tissue blood vessels with a pro-apoptotic motif. Studied in animal obesity models, its clinical development was discontinued. It is not an approved therapeutic in Canada.

Source & context

Biological / chemical source: Synthetic peptidomimetic combining a homing sequence with a pro-apoptotic motif

Primary research or clinical context: Discontinued investigational compound; not an approved drug in Canada

Targeting design

Adipotide is built as a chimeric molecule: a peptide that homes to prohibitin and ANXA2 on the blood vessels feeding white adipose tissue, fused to a KLAKLAK-type sequence intended to disrupt mitochondrial membranes. The concept was to reduce fat depots by targeting their vascular supply.

Research findings and limits

Published work reported rapid weight loss in mouse and non-human-primate models, generating substantial interest. However, development was discontinued, and human efficacy and safety were not established. Search popularity outpaces the strength of clinical evidence for this compound.
