GHRP-1 was among the earliest peptidyl growth hormone secretagogues synthesized after the discovery that enkephalin-amide analogs could release GH. It is closely related to GHRP-2, GHRP-6 and hexarelin, sharing the ghrelin-receptor mechanism rather than any sequence homology with GHRH.
